A cofactor for factor XI activation.

چکیده
mice carry mutations of the c-Kit gene, which obviously do not affect basophil biology. On the other hand, basophil-deficient Mcpt8DTR or basoph8 mice carry a transgenic receptor that mediates toxin-mediated cell death and mast cells in these mice remain intact. In mice deficient in IL-3, both mast cell and basophil generation are severely impaired only after parasite infection.6 Therefore, Cpa3-Cre; Mcl-1fl/fl mice will prove a useful animal model from which both redundant and nonredundant functions of mast cells and basophils in vivo can be unveiled. Polyphosphate is a cofactor for the activation of factor XI by thrombin.
Factor XI deficiency is associated with a bleeding diathesis, but factor XII deficiency is not, indicating that, in normal hemostasis, factor XI must be activated in vivo by a protease other than factor XIIa. Several groups have identified thrombin as the most likely activator of factor XI, although this reaction is slow in solution. Although certain nonphysiologic anionic polymers and surfaces...
متن کاملActivated factor V is a cofactor for the activation of factor XI by thrombin in plasma.
The mechanism by which the intrinsic pathway of coagulation contributes to physiological hemostasis is enigmatic. Thrombin activates factor XI, a key zymogen in this pathway, which leads to increased thrombin generation. As thrombin-dependent activation of factor XI in vitro is relatively inefficient, we hypothesized that a physiological cofactor supports this reaction in a plasma environment. ...
متن کاملRole of high-molecular-weight kininogen in surface-binding and activation of coagulation Factor XI and prekallikrein.
In the contact phase of activation of the kinin-forming, intrinsic clotting, and fibrinolytic systems, high-molecular-weight kininogen acts as a cofactor for the activation of Factor XI, prekallikrein, and Hageman factor. One mechanism by which high-molecular-weight kininogen acts as a cofactor has been studied by using 125I-labeled Factor XI and prekallikrein in kaolin-activated normal human p...
متن کاملThrombin activates factor XI on activated platelets in the absence of factor XII.
Thrombin can activate factor XI in the presence of dextran sulfate or sulfatides. However, a physiological cofactor for thrombin activation of factor XI has not been identified. We examined this question in a cell-based, tissue factor-initiated model system. In the absence of factor XII, factor XI enhanced thrombin generation in this model. The effect on thrombin generation was reproduced by 2 ...
متن کاملFeedback Activation of Factor XI by Thrombin in Plasma Results in Additional Formation of Thrombin That Protects Fibrin Clots From Fibrinolysis
Recently, an alternative pathway for factor XI activation has been described in which factor XI is activated by thrombin. Patients with a factor XI deficiency bleed mostly from tissues with high local fibrinolytic activity. Therefore, the role of thrombin-mediated factor XI activation in both fibrin formation and fibrinolysis was studied in a plasma system. clotting was induced by the addition ...
متن کاملPlatelet-Derived Short-Chain Polyphosphates Enhance the Inactivation of Tissue Factor Pathway Inhibitor by Activated Coagulation Factor XI
INTRODUCTION Factor (F) XI supports both normal human hemostasis and pathological thrombosis. Activated FXI (FXIa) promotes thrombin generation by enzymatic activation of FXI, FIX, FX, and FV, and inactivation of alpha tissue factor pathway inhibitor (TFPIα), in vitro. Some of these reactions are now known to be enhanced by short-chain polyphosphates (SCP) derived from activated platelets. Thes...
